ABSTRACT

Abstract In drug therapy, it is important to provide therapeutic levels of drug to the site of action and maintain them during the treatment. This work describes the in vitro release of alendronate from sodium alginate cross-linked Montmorillonite (MMT) composite beads. Effect of crosslinking cation, concentration of montmorillonite and media on encapsulation efficiencies, and release profiles of alendronate were studied. Beads were characterized using equilibrium swelling ability study, Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), Energy-dispersive x-ray spectroscopy (EDX) and scanning electron microscopy (SEM). Results indicate that addition of montmorillonite increases the encapsulation efficiencies and slows down the release rates significantly.

ABSTRACT

OBJECTIVE:To investigate the effects of alendronate sodium on postoperative indexes in the osteoporosis patients with thoracolumbar fracture. METHODS :Totally 170 patients with osteoporotic thoracolumbar fracture admitted in 416 Hospital of Nuclear Industry during Jan.-Dec. 2018 were divided into control group (85 cases)and observation group (85 cases)according to random number table . All patients underwent percutaneous vertebroplasty or percutaneous kyphoplasty. After operation ,control group received symptomatic and supportive treatment. Observation group was additionally given Alendronate sodium tablets 70 mg orally,once a week ,on the basis of control group. Treatment course of 2 groups lasted for one year. VAS score ,bone metabolism indexes [serum phosphorus (P), calcium (Ca), osteocalcin (OST), bone alkaline phosphatase (BAP)], contents of osteoprotegerin(OPG)and nuclear factor-κB receptor activator ligand(RANKL),cytokines(TNF-α,IFN-γ,IL-10)levels were observed in 2 groups before and after 1,3,6,12 months of treatment,and ADR were recorded . RESULTS :Before treatment , there was no significant difference in VAS score ,bone metabolism indexes ,contents of OPG and RANKL and cytokines levels between 2 groups (P>0.05). After treatment ,VAS scores of 2 groups were significantly lower than before treatment ,the observation group was significantly lower than the control group after 1,3,6 months of treatment (P<0.05). OST and BAP contents of observation group after 3,6,12 months treatment as well as OPG and RANKL contents ,TNF-α and IFN-γ levels after 1,3,6,12 months of treatment were significantly lower than before treatment and control group at the same period ;IL-10 level was significantly was higher than before treatment and control group at the same period (P<0.05). There was no statistical significance in P or Ca of 2 groups before and after treatment ,OST,BAP,OPG,RANKL contents and cytokines levels of control group before and after treatment (P>0.05). There was no statistical significance in total incidence of ADR between 2 group(P> 0.05). CONCLUSIONS :Alendronate sodium can significantly relieve the pain and improve cytokines ,bone metabolism indexes , and inhibit expression of releated pathways in osteoporosis patients with thoracolumbar fracture ,with good safety.

ABSTRACT

OBJECTIVE:To study therapeutic efficacy of total hip arthroplasty(THA)combined with Alendronate sodium tablets in the treatment of femoral neck fracture and its effects on bone mineral density(BMD). METHODS:A total of 98 patients with femoral neck fractures in our hospital during 2014-2016 were divided into observation group and control group by random digital table method,with 49 cases in each group. Both groups were treated with THA. 7 days after operation,control group was given routine anti-osteoporosis treatment [Gaierqi D tablets(containing 600 mg calcium/vitamin D3125 IU in each tablet),p.o., one tablet/time,once a day;Calcitriol soft capsules(25 μ g each pill,p.o.,2 pills/time,once a day)]. Observation group was additionally given Alendronate sodium tablets(10 mg/time,once a day)on the basis of control group. Both groups were treated for consecutive 3 months. The hip function excellent rate was evaluated by using Harris scoring system at 7 days,3 months and 6 months after operation. The periprosthetic 7 cases of egion interest(ROI1-7)BMD were detected and compared between 2 groups. RESULTS:The excellent rate of hip joint function in the control group were 16.33%,40.82%,69.39% 7 days,3 months,6 months after operation,respectively;those of observation group were 17.78%,73.33%,88.89% respectively. There was no statistical significance in the excellent rates of hip joint function between 2 groups 7 days after operation(P>0.05). The excellent rate of hip joint function in observation group was higher than control group 3 months and 6 months after operation(P<0.05). There was no significant difference in periprosthetic BMD between 2 groups 7 days after operation(P>0.05). Compared with 7 days after operation,BMD of ROI1,ROI6 and ROI7 in 2 groups were decreased gradually 3 months and 6 months after operation (P<0.05 or P<0.01). BMD of ROI2,ROI3 and ROI5 decreased first and then increased(P<0.05 or P<0.01). There was no significant change in BMD of ROI4(P>0.05). There was no significant difference in BMD of each area between 2 groups 3 months after operation(P>0.05). BMD of ROI1,ROI2,ROI3,ROI5,ROI6 and ROI7 in observation group were significantly higher than control group 6 months after operation(P<0.05 or P<0.01). CONCLUSIONS:THA combined with Alendronate sodium tablets and conventional anti-osteoporosis treatment can improve the excellent rate of hip joint function in patients with femoral neck fracture and the level of periprosthetic BMD.

ABSTRACT

<p><b>OBJECTIVE</b>To compare the efficacy differences between catgut implantation at stellate ganglion combined with oral administration of alendronate sodium and oral administration of alendronate sodium alone on postmenopausal osteoporosis (PO).</p><p><b>METHODS</b>Sixty patients of PO were randomly divided into an observation group and a control group, 30 cases in each one. The patients in the control group were treated with oral administration of alendronate sodium. Based on the treatment of control group, the patients in the observation group were treated with catgut implantation at stellate ganglion. The treatment was given once a week in the two groups; the consecution treatment of four weeks constituted one session, and totally six sessions were given. The changes of total syndrome score, bone mineral density of lumbar vertebra (L to L) and femeral neck (FN) and estradiol were observed before and after treatment; the clinical efficacy was compared between the two groups.</p><p><b>RESULTS</b>Compared before treatment, the total syndrome score, bone mineral density of lumbar vertebra (L to L) and FN and estradiol were significantly improved after treatment (all <0.05); which were more significant in the observation group (all <0.05). Compared before treatment, the level of estradiol in the control group was not significantly changed after treatment (>0.05), while that in the observation group was significantly changed after treatment (<0.05). After treatment, the level of estradiol in the observation group was higher than that in the control group (<0.05). The total effective rate was 93.3% (28/30) in the observation group, which was significantly higher than 83.3% (25/30) in the control group (<0.05).</p><p><b>CONCLUSION</b>Catgut implantation at stellate ganglion combined with oral administration of alendronate sodium are superior to oral administration of alendronate sodium alone for postmenopausal osteoporosis, which improve the clinical symptoms, regulate the hormone level and increase bone mineral density.</p>

ABSTRACT

Objective To observe the clinical effect of psychological intervention combined with alendronate and low molecular heparin calcium on the treatment of senile osteoporotic fracture. Methods From February 2014 to March 2017, clinical data of 82 elderly patients with osteoporotic fracture in the first people's hospital in Jiande were retrospectively analyzed. All the patients were randomly divided into the control group and the experimental group with 41 cases in each group. The control group patients were given alendronate sodium and low molecular heparin calcium; At this basis, the experimental group were received psychological nursing intervention. The effect in the two groups was compared. Results After treatment, bone mineral density (BMD) index and pain scores in the two groups were significantly improved. BMD and pain scores in the experimental group were significantly better than those in the control group (P<0.05). The total effective rate was 95.12% (39/41) and the complication rate was 17.07% (7/41) in the experimental group, while in the control group were respectively 70.73% (29/41), 36.59% (15/41), the differences were statistically significant (P<0.05). Conclusion Psychological intervention combined with alendronate sodium and low molecular heparin on the treatment of senile osteoporotic fracture has significant effect, can effectively improve the clinical symptoms of patients, reduce the incidence of complications, with clinical application value.

ABSTRACT

Objective To evaluate the efficacy of alendronate sodium combined with dietary and exercise intervention in postmenopausal osteoporosis. Methods 88 postmenopausal osteoporosis patients were enrolled and treated with alendronate sodium and Vitamin D combined with dietary and exercise intervention for 12 months. Bone mineral density (BMD) of lumber spine and femur neck were measured before and after the treatment. Results Medication combined with dietary and exercise intervention significantly increased bone mineral density of postmenopausal osteoporosis patients, and the serum levels of bone alkaline phosphatase (BLAP), alkaline phosphatase (AKP) and tartrate resistant acid phosphatase (TRACP) were significantly higher than those pre-treatment, with significant differences (P<0.05). Conclusion Compared to simple medication treatment, medication combined with dietary and exercise intervention is more effective to enhance medication efficacy and BMD level.

ABSTRACT

OBJECTIVE:To evaluate the economics of alendronate sodium preventing osteoporotic fractures in postmenopausal women,and to provide reference for osteoporotic and osteopenia patients to select economical and effective therapy plan. METH-ODS:Using Markov economic model,TreeAge Pro 2011 software was used for cost-utility analysis using RCTs literature data of basic medication(calcium and vitamin D3)and basic medication combined with alendronate sodium as baseline data. Using quality adjusted life years(QALYs)and incremental cost utility ratio(ICUR)as economics evaluation indexes,the stability of established model was investigate by single factor sensitivity analysis. RESULTS:QALYs of the basic treatment group was 0.704 years,and QALYs of drug combination group was only 0.708 years. However,ICUR of two groups reached 714252.44. For the late period of hip fracture and other fractures,drug combination group was in high cost and low benefit,obviously had no economic advan-tage. For vertebral fractures,the cumulative probability of fracture in drug combination group was significantly lower than basic treatment group,and ICUR of the two groups was only 13902.17,with economic advantage. For hip fracture,the cumulative prob-ability of fracture in basic treatment group was the same as drug combination group,and ICUR was 19109.00,with economic ad-vantage. For wrist fractures,curative effect and economy needed further study. CONCLUSIONS:For the low-risk population with osteoporotic fractures,calcium and vitamin D3 are more economical in preventing osteoporosis fractures. For the high-risk popula-tion with vertebral and hip fractures,alendronate sodium combined with basic treatment is effective and economical.

Introduction: Different rates of resorption are mediated by osteoclasts that may be affected by bisphosphonates during bone graft repair. Bisphosphonates are drugs that act as inhibitors of bone resorption. Objective:The aim of the present study was to evaluate the rate of resorption of skullcap grafts in rabbits with and without the use of alendronate sodium. Material and method: Thirty two New Zealand rabbits were divided into two groups (control group and alendronate group) and divided again into four periods (7, 14, 30 and 60 days). The control group did not receive alendronate, while animals of the experimental group received 4 mg of alendronate sodium weekly after the surgery. An 8 mm diameter bone block was removed from the parietal bone and fixed by screws to the contralateral parietal bone. During the periods of 7, 14, 30 and 60 days, the animals had undergone euthanasia and samples were removed for further analysis. Morphological and histomorphometric tests were used to compare graft thicknesses and to evaluate the newly formed bone at the interface between the graft and receptor site. The Wilcoxon and Mann-Whitney tests were used for statistical analyses. Result: All grafts healed and integrated uneventfully and no statistically significant differences in resorption rates or bone deposition were detected after the final incorporation of the graft in both groups. Conclusion: Alendronate Sodium did not decrease the bone graft resorption rates, but there was a tendency for better results in the control group regarding the resorption and neoformation in autogenous calvarial bone grafts in rabbits.


Introdução: Durante o reparo de enxertos ósseos, diferentes taxas de reabsorção são mediadas pelos osteoclastos e podem ser afetadas pelos bisfosfonatos, que são drogas que agem como inibidores da reabsorção óssea. Objetivo: Avaliar a taxa de reabsorção dos enxertos ósseos de calota craniana de coelhos, com e sem o uso alendronato de sódio. Material e método: Trinta e dois coelhos Nova Zelândia foram divididos igualmente em 2 grupos (grupo controle e grupo alendronato de sódio) e subdivididos em 4 períodos (7, 14, 30 e 60 dias). O grupo controle não recebeu alendronato, enquanto os animais do grupo experimental receberam 4 mg de alendronato de sódio por semana, em dose única, após a cirurgia. Um bloco de osso de diâmetro de 8 mm foi retirado o osso parietal e fixado com parafuso no osso parietal contralateral. Após cirurgia, nos períodos de 7, 14, 30 e 60 dias, os animais foram eutanasiados e as peças removidas para análise. Análises morfológica e histomorfométrica foram utilizados para comparar a espessura do enxerto e para avaliar a interface de osso recém formado entre o enxerto ósseo e o sítio receptor. Os testes de Wilcoxon e Mann-Whitney foram utilizados para as análises estatísticas. Resultado: Todos os enxertos repararam e integraram sem intercorrências; não foram detectadas diferenças estatisticamente significativas nas taxas de reabsorção ou deposição óssea, após a incorporação final do enxerto em ambos os grupos. Conclusão: Alendronato de sódio parece não diminuir a taxa de reabsorção, porém houve uma tendência de resultados melhores no grupo controle tanto na reabsorção quanto na neoformação óssea em enxertos ósseos autógenos de calota craniana de coelhos.

ABSTRACT

As a mono-sodium salt form of alendronic acid, alendronate sodium presents multi-level ionization for the dissociation of its four hydroxyl groups. The dissociation constants of alendronate sodium were determined in this work by studying the piecewise linear relationship between volume of titrant and pH value based on acid-base potentiometric titration reaction. The distribution curves of alendronate sodium were drawn according to the determined pKa values. There were 4 dissociation constants (pKa1=2.43, pKa2=7.55, pKa3=10.80, pKa4=11.99, respectively) of alendronate sodium, and 12 existing forms, of which 4 could be ignored, existing in different pH environments.

ABSTRACT

OBJECTIVE:To explore the effects of alendronate sodium on bone mineral density (BMD) and bone marker in prostate cancer elderly patients after medical castration therapy. METHODS:In perspective study,84 elderly patients undergoing medical castration therapy were selected and divided into treatment group(45 cases)and control group(39 cases)according to ran-dom number table. Control group received medical castration therapy+Calcium carbonate D3 tablets,1 tablet,po,qn;treatment group was additionally given Alendronate sodium tablets 70 mg,po,once a week,1 week after routine treatment,on the basis of control group. Treatment course of 2 groups lasted for 12 months. The levels of 25-OH-D,testosterone,BMD and bone marker were observed in 2 groups,and the occurrence of ADR was recorded. RESULTS:3 cases of treatment group and 1 case of control group dropped out of the study. Before treatment,there was no statistical significance in above indexes between 2 groups (P>0.05). After treatment,25-OH-D levels of 2 groups were increased slightly,but there was no statistical significance(P>0.05);tes-tosterone level of 2 groups were decreased significantly compared to before treatment,with statistical significance (P0.05). CONCLUSIONS:Alendronate sodium can prevent bone loss and reduce the rate of bone turnover in elderly patients with prostate cancer receiving medical castration therapy.

ABSTRACT

OBJECTIVE:To observe clinical efficacy and safety of Xianlinggubao combined with alendronate sodium in the treatment of osteoporosis. METHODS:112 patients with osteoporosis were randomly divided into treatment group and control group with 56 cases in each group. Both group were given routine treatment as calcium carbonate,vitamin D and life style. Control group were given alendronate sodium 10 mg orally,qd;treatment group was additionally given Xianlinggubao capsule 1.5 g,bid, on the basis of control group. Both groups received 12 weeks of treatment. The levels of S-Ca,S-P,24 h urinary calcium(U-Ca), alkaline phosphatase(ALP),osteocalcin,IL-6,bone mineral density(BMD)of L2-4 lumbar spine and femoral neck were observed in 2 groups before and after treatment. Clinical efficacy and toxic reaction were compared. RESULTS:There was no statistical sig-nificance in the levels of S-P and S-Ca between 2 groups after treatment(P>0.05);the level of ALP、osteocalcin、BMD increased in treatment group after treatment,the level of U-Ca and IL-6 decreased compared to before treatment,and the treatment group was better than the control group,with statistical significance(P<0.05). Total effective rate of treatment group(92.86%)was sig-nificantly higher than that of control group (73.21%),with statistical significance (P<0.05). No obvious toxic side effects was found in 2 groups. CONCLUSIONS:Xianlinggubao combined with alendronate sodium shows significant therapeutic efficacy for patients with osteoporosis with good safety.

ABSTRACT

OBJECTIVE:To investigate clinical efficacy and safety of conjugeted estrogen tablets combined with Salmon calci-tonin acetate injection and Alendronate sodium tablets in the treatment of osteoporosis in postmenopausal. METHODS:148 post-menopausal women with osteoporosis were randomly divided into observation group(78 cases)and control group(70 cases). Con-trol group was given Salmon calcitonin acetate injection hypodermically or intramuscularly,100 IU each time,once a day in first week,every two days in second week,every three days in third week,21 times of consecutive injection as a treatment course;Alendronate sodium tablet,70 mg each time,once a week,3 months as a treatment course. Based on control group,observation group was additionally given conjugeted estrogen tablets,0.625 mg each time,qd,for consecutive 3 weeks,drug withdrawal for one week,and then continued to taking drug,for 3 months in total. Clinical efficacy of 2 groups were observed as well as BGP, bone metabolism index as human collagen type Ⅰ N-terminal peptide (NTX) and ALP,bone density and ADR before and after treatment. RESULTS:After treatment,total effective rate of observation group was 94.87%,which was significantly higher than 78.57% in control group,with statistical significance(P0.05);various ADR disappeared after drug withdrawal. CONCLUSIONS:Conjugeted estrogen tablets combined with Salmon calcitonin acetate injection and Alendronate sodium tablets can effectively relieve bone pain,regulate bone metabolism,increase bone density and induce slight ADR in postmenopausal with osteoporosis.

ABSTRACT

Objective To investigate the clinical effect of small incision for total hip arthroplasty on osteoporosis treatment of elder patients with fracture of the femoral neck.Methods Fifty-six elder patients with femoral neck methods were selected as our subjects,who were hospitalize from Jan.2011 to Dec.2013 in Xiahuayuan Meikuang Hospital,Zhangjiakou Company Limited.They were randomly divided into the observation group (26 cases) and control group (30 cases).All patients were treated with small incision total hip arthroplasty.The patients in the observation group were given the alendronate sodium.The curative effect,hospitalization time,bone mineral density were recorded.Results The long-term efficacy of the excellent and good rate of the observation group was 96.2% (25/26),higher than that in control group(86.6% (26/30)),and the difference was statistically significant(x2 =4.125,P < 0.05).The length of hospitalized time in observation group was (26 ± 4) d,shorter than that in the control group (35 ± 3) d,and the difference was statistically significant(t =9.102,P < 0.05).The bone density of patients in the observation group was (0.64 ± 0.03) g/cm2,higher than that in control((0.61 ± 0.05) g/cm2),and there was significant difference between two groups (t =8.461,P < 0.05).Conclusion The effect of small incision total hip arthroplasty combined with anti osteoporosis drugs for the treatment of femoral neck fractures in the elderly is good and worthy of clinical usage.

Objective To observe the effects of alendronate sodium and alfacalcidol on the parameters of atherosclerosis in old women with type 2 diabetes mellitus(T2DM ) complicating osteoporosis .Methods 28 old women with T2DM complicating osteopo-rosis were taken as the osteoporosis group and 25 old women with T2DM and without osteoporosis were taken as the control group .Alendronate sodium(70 mg/week) and alfacalcidol(0 .5 μg/d) were given to the osteoporosis group for 12 month .The pa-rameters of the bone mass density (BMD)of femoral neck and ,T value ,arkle brachial index (ABI) ,cardio-ankle vascular index (CAVI) and carotid artery intima-media thickness(IMT) ,blood fat and glycosylated hemoglobin were observed .Results Com-pared with the control group ,BMD ,T value and ABI in the osteoporosis group were significantly decreased (P<0 .01) ,while CAVI and IMT were obviously increased(P<0 .05) .Compared with before treatment ABI ,CAVI and IMT after 12 month treatment in the osteoporosis group had no significant changes ,but BMD and T value of femoral neck and ward triangle area were significantly increased(P<0 .05) ,the atherosclerosis degree after 12 months in the control group was significantly aggravated (P<0 .05) .BMD and T value had no obvious change .Conclusion Alendronate sodium combined with alfacalcidol for treating T 2DM complicating os-teoporosis can improve the atherosclerosis parameters and has certain role in preventing and treating cardiovascular and cerebrovas-cular diseases .

ABSTRACT

Objective Toobserve the effect of exercise combined with alendronate sodium tablets (Ain) on receptor activator of nuclear factor kappa B ligand (RANKL)/osteoprotegerin (OPG) expression and mitogen-activated protein kinase (MAPK) signaling pathways in ovariectomized (OVX) rats. Methods A total of 90 6-month-old female SDrats were evenly randomized into sham group, OVX group, Ain treatment (OVX+Aln) group, exercise therapy (OVX+EX) group, and Ain combined with exercise treatment (OVX+Aln+EX) group. Eight weeks after ovariectomy, rats in the latter three groups were treated with intragastric 1 mg/(kg • d) Ain and/or exercise. After 12 weeks of treatment, dual-energy X-ray absorptiometry was used to examine the bone mineral density (BMD) of the left and right femurs. Enzyme-linked immunosorbent assay (ELISA) was used to determine the serum levels of RANKL and OPG. Real-time PCR was used to examine RANKL and OPG mRNA expression in bone tissues. Western blotting analysis was used to examine the phosphorylation of c-Jun N-terminal kinase (JNK) » p38 MAPK (p38), extracellular-regulated kinase (ERK), and c-Fos protein expression in bone tissues. Results The following resultswere observed after 12-week treatment. (1) Compared with sham group, the BMD values of the lett and right femurs were significantly decreased in OVX group (P<0. 05). The serum RANKL level was significantly increased, serum OPG level was significantly decreased, and the RANKL/OPG ratio was significantly increased (all P<0. 05). The RANKL mRNA expression was significantly increased and OPG mRNA expression was significantly decreased in bone tissues (all P<0. 05). (2) ComparedwithOVX group, the BMD values of left and right femurs of OVX+Aln, OVX+EX and OVX+Aln+EX groups were significantly increased (all P<0. 05). The serum RANKL level was significantly decreased, serum OPG level was significantly increased, and the RANKL/OPG ratio decreased (all P<0. 05). The RANKL mRNA expression was significantly decreased, and OPG mRNA expression was significantly increased in bone tissues (all P<0. 05). The femoral BMD values, serum RANKL and OPG levels, and RANKL and OPG mRNA expression in bone tissues in OVX+Aln+EX group were significantly different from those in OVX+Aln group and OVX+EX group (all P<0. 05). (4) Compared with sham group, bone tissue p-JNKand c-Fos expression in OVX group was significantly increased (all P<0. 05). However, compared withOVX group, bone tissue p-JNK and c-Fos expression in OVX+Aln, OVX+EX and OVX+Aln+EX groups was significantly decreased (all P<0. 05), and there were no significant differences among the latter three groups. p-p38 and p-ERK expression was similar in all groups. Conclusion Exercise combined with Ain can improve the BMD values of left and right femurs in OVX rats, which may be related to the regulation of RANKL/OPG balance and inhibition of JNK phosphorylation and c-Fos expression.

ABSTRACT

Objective To investigate the clinical effect of alendronate sodium on bone metabolism and bone mineral density (BMD) for treating the elderly female with osteoporotic distal radius fractures.Methods Ninety elderly female with osteoporotic distal radius fractures patients were divided into treatment group and control group with 45 cases each according to the method of random number list.The two groups were given the medication treatment after 3 d of external fixation stenting.The control group was given calcium dimension D and the treatment group was given alendronate sodium and calcium dimension D.The course of treatment was 6 months.Before and after treatment,the BMD and bone metabolism [bone alkaline pbosphatase(BALP),tartrate-resistant acid phosphatase-5b(TRACP-5b)] were detected in both groups.The wrist function of two groups was evaluated.Results Before treatment,the level of BMD of treatment group and control group were (0.472 ± 0.157),(0.519 ± 0.173) g/cm2,there was no significant difference (P >0.05).After treatment,the level of BMD of treatment group and control group were (0.893 ±0.298),(0.571 ± 0.184) g/cm2,the BMD of treatment group after treatment was significantly higher than that before treatment and control group after treatment(P < 0.05).The level of BMD of control group after treatment had no significant difference compared with before treatment (P > 0.05).Before treatment,the level of BALP,TRACP-5b of treatment group and control group were (53.76 ± 17.92),(5.61 ± 1.87) U/L and (51.86 ± 17.29),(5.54 ± 1.85) U/L,the level of BALP,TRACP-5b after treatment had significant difference compared with before treatment and control group after treatment (P <0.05).There was no significant difference in control group before treatment and after treatment (P > 0.05).The good rate of rehabilitation of wrist function in treatment group was 91.1% (41/45),significantly higher than that in control group [51.1% (23/45)] (x2 =17.524,P <0.01).Conclusions The alendronate sodium has a role to suppress bone resorption and increase BMD.It can promote the recovery of wrist function.

Resultados: Houve movimentação significante após 5 e 7 dias (P<0.05) em todos os grupos em comparação aos lados sem movimentação. Entretanto, no grupo ovariectomizado houve maior quantidade de movimentação dentária induzida comparado aos outros grupos, com resultado estatisticamente significante somente aos 7 dias. Conclusão: O alendronato de sódio na dosagem empregada diminuiu a quantidade de movimentação dentária induzida em comparação ao grupo OVX, sendo similar ao grupo controle.

Conclusion: Alendronate sodium dosage employed decreased the amount of induced tooth movement compared to the OVX group, being similar to the control group.

ABSTRACT

OBJETIVO: Avaliar resultados do uso de alendronato de sódio em pós operatório de fratura transtrocanteriana. MÉTODO: Durante seis meses, 75 pacientes foram atendidos no Serviço de Ortopedia devido à fratura transtrocanteriana de fêmur, sendo submetidos a tratamento cirúrgico com fixação através de placa DHS 135º. Foram selecionados 19 pacientes, hígidos, cooperativos, previamente deambuladores, classificados segundo Tronzo em tipo III e cujos quadris contra laterais não haviam sido submetidos a nenhuma cirurgia prévia, permitindo a realização de Densitometria Óssea Mineral no pós-operatório imediato e após seis meses. Os pacientes foram divididos em grupos I e II, com e sem uso de alendronato de sódio, a partir do pós-operatório imediato, respectivamente. O seguimento mínimo foi de seis meses. Após quatro semanas, os pacientes foram avaliados de acordo com a Escala Visual Analógica de Dor (EVA), para observação do índice de reabsorção óssea, a massa óssea e a consolidação radiológica da fratura. RESULTADOS: Houve uma tendência estatística de consolidação radiológica mais precoce com menor reabsorção óssea nos pacientes com uso de alendronato de sódio na pós-fratura com osteoporose estabelecida. CONCLUSÃO: Concluímos que, radiologicamente, os pacientes que utilizaram alendronato de sódio tiveram uma consolidação óssea mais rápida, com menor índice de reabsorção.


OBJECTIVE: To evaluate the results of the use of alendronate sodium in postoperative transtrochanteric femoral fractures. METHOD: Over a six-month period, 75 patients were treated at the Orthopedic Service for transtrochanteric femoral fractures, undergoing surgery with 135 degree DHS plate fixation. We selected 19 patients who were healthy, cooperative, previously unable to walk, classified as type III according to Tronzo, whose contralateral hips had not undergone any previous surgery, enabling the evaluation of bone mineral density (BMD) in the immediate postoperative period and after six months. The patients were divided into groups I and II, with and without the use of alendronate sodium in the immediate postoperative period, respectively. The minimum follow-up was six months. After four weeks, the patients were evaluated according to the Visual Analogue Scale for Pain (VAS) in order to verify the rate of bone resorption, bone mass and radiological consolidation of the fracture. RESULTS: There was a statistical trend of earlier radiological consolidation and lower bone resorption in patients with the use of alendronate sodium in post-fracture with established osteoporosis. CONCLUSION: We concluded that radiologically, patients using alendronate sodium showed faster bone consolidation, with a lower rate of resorption.

ABSTRACT

@#Objective To observe the effect of progressive resistance exercise combined with alendronate sodium on bone mineral density(BMD)of lumbar spine in patients with postmenopausal osteoporosis.Methods 20 patients with postmenopausal osteoporosis were randomly divided into the A and B group with 10 cases in each group.The cases of the group A were treated with progressive resistance exercises combined with alendronate sodium.Those of the group B only took alendronate sodium orally.The course of two groups was 3 months.BMD of lumbar spine was measured by dual-energy X-ray absorptiometry before and 3 months after treatment.Results Before treatment,BMD of two groups was not different.After 3 months treatment,BMD of lumbar spine were significantly improved in group A(raised 4.520±0.68%)than group B(raised 0.100±0.01%),there was a significant difference between two groups(P<0.01).Conclusion Progressive resistance exercises combined with alendronate is more efficacious than alendronate alone in restoring lumbar spine BMD in patients with postmenopausal osteoporosis.
